Frequently Asked Questions
These terms describe different filtration objectives. Clarification generally addresses suspended solids or particulates. Bioburden-reduction filtration aims to reduce microbial load before a later process step. Sterilizing-grade filtration is used where microbial retention is a critical process objective and requires process-specific selection, operating conditions, and integrity-testing considerations. One stage does not automatically substitute for another.
Filter integrity testing is a non-destructive test used to assess whether a wetted membrane filter performs within established acceptance criteria. Methods can include bubble point, diffusive flow, forward flow, or pressure-hold testing, depending on the filter, test method, and validated procedure. An integrity test does not, by itself, demonstrate product sterility.
PUPSIT means Pre-Use, Post-Sterilization Integrity Testing. It is performed after the filter assembly has been sterilized and before product processing begins. It can help confirm filter and assembly integrity before use, but its implementation must account for wetting, pressure limits, air management, drainage, flow-path design, and the site’s documented process controls.
PUPSIT is not expressly required by U.S. cGMP regulations in the same way it is addressed in EU GMP Annex 1 expectations. However, individual requirements can depend on the product, market, quality system, contamination-control strategy, and documented risk assessment. Regulatory and validation decisions should be reviewed by the site’s quality and technical stakeholders.
See our PUPSIT Readiness Playbook for more information about how to make your US operation PUPSIT-ready.
In normal-flow filtration, fluid moves toward the filter media and the filtered stream passes through it. Retained material can accumulate on the filter surface or within the filter structure. In tangential-flow filtration, fluid flows parallel to the membrane surface while a portion passes through as permeate. TFF is commonly considered for concentration, diafiltration, and applications where continuous crossflow can help manage retained material.
A meaningful evaluation typically starts with the filtration objective, fluid or product category, process stage, anticipated volume, solids or contaminant characteristics, operating conditions, current filtration setup, and relevant documentation needs. The information needed will vary depending on whether the goal is clarification, microbial control, concentration, diafiltration, dispensing, or single-use fluid handling.
